Protein function and interaction

JARID1 proteins are histone demethylases that belong to the ARID family of DNA binding proteins and act as transcriptional repressors. PLU-1/JARID1B specifically removes methyl groups from di- and trimethylated histone 3 lysine 4 (H3K4) and presents a restricted pattern of expression (Yamame et al., 2007 PMID: 17363312; Secombe & Eisenman, 2007 PMID: 17568193; Xiang et al., 2007 PMID: 18048344; Christensen et al., 2007 PMID: 17320161).

A yeast two-hybrid screen using PLU-1 as bait has identified two interacting proteins, brain factor-1 (BF-1) and paired box 9 (PAX9), both of which are developmental transcription factors (Tan et al., 2003 PMID: 12657635). Precipitation of AR (androgen receptor) was able to pull down JARID1B when both proteins were coexpressed (Xiang et al., 2007 PMID: 18048344). Co-immunoprecipitation assays provided an evidence that JARID1B (RBP2-H1) can bind to RB-1 (Roesch et al., 2005 PMID:15803180). Chromatin immunoprecipitation assays confirmed that PLU-1/JARID1B can interact with the promoters of the metallothionein genes (Scibetta et al., 2007 PMID: 17709396) and with the promoter of tumor suppressor genes, including BRCA1 (Yamame et al., 2007 PMID: 17363312). A phage display screen using JARID1B as bait identified hPc2 of the Polycomb group of protein (Zhou et al., 2009 PMID: 19336002).

JARID1B plays a role in the regulation of mouse neural cell fate and differentiation (Dey et al., 2008 PMID: 18591252).
