ADAM29 contains all the domains of a typical ADAM molecule, including a signal peptide, a proprotein domain, a metalloprotease domain, a disintegrin domain, a cysteine-rich domain, a transmembrane region, and a cytoplasmic tail. Within the cysteine-rich domain, a short hydrophobic stretch can be modeled into an amphipathic helix with a central proline residue, a characteristic of the putative fusion peptide identified in the membrane-anchored subunit, suggesting that ADAM29 may play a role in mediating cell–cell fusion (Xu et al., 1999 PMID: 10644455).
