Protein function and interaction
CAGE promotes motility of cancer cells through activation of Focal adhesion kinase, FAK (Shim et al, 2006a PMID:17028776) and by activating ERK and p38 MAPK, inducing catalase activity, and reducing ROS levels (Shim et al., 2006b PMID:16819299).
CAGE expression is cell cycle related, suggesting that it plays a role in cellular growth and proliferation (Cho et al., 2002 PMID: 11922625). CAGE possesses oncogenic potential and promotes cell cycle progression by inducing AP-1- and E2F-dependent expression of cyclin D1 and E (Por et al., 2010 PMID: 20220142).
CAGE regulates the expression of epithelial-mesenchymal transition (EMT)-related proteins through ERK, Akt and nuclear factor kappaB (NF-kB). CAGE also induces c-FLIP(L) and Snail to enhance cell motility and increase resistance to anti-cancer drugs (by similarity) (Kim et al., 2009 PMID: 19322522) and induces the expression of matrix metalloproteinase-2 (MMP-2) by activating Akt, which in turn interacts with inhibitory kappa kinase beta (IkappaKbeta) to activate nuclear factor kappaB (NF-kappaB) (Kim & Jeong, 2009 PMID: 19944019).
