Gene Identification
MAGEA1 was the first molecularly characterized human tumor antigen recognized by cytotoxic T cells. Initially, a MAGEA-1 antigen, denominated E antigen, was identified by selecting sub clones from the MZ2-MEL cell line, derived from a melanoma patient, which are resistent to lysis by autologous lymphocyte derived CTLs (Van den Eynde et al, 1989 PMID: 2529220). Afterwards, the gene encoding antigen E was identified by T cell epitope cloning (van der Bruggen et al., 1991 PMID: 1840703; Traversari et al., 1992 PMID: 1402688; Coulie et al, 1993 PMID: 8280701).
Gene structure and chromosomal localization
MAGEA-1 was mapped to the Xq28 region by in situ hybridization of metaphase spreads using a MAGEA-1 probe (Wang et al, 1994 PMID: 8039421).
